چکیده

Background A specific feature of the autoimmune response in rheumatoid arthritis (RA) is presence anti-citrullinated protein antibodies (ACPAs) patient sera. These can appear several years before disease onset and are involved development RA. Objective We performed proteomic analysis by mass spectroscopy to identify novel citrullinated antigens autoantibodies RA patients. Methods Polypeptides isolated from sera patients were identified Orbitrap high-precision spectrometry then citrulline-containing proteins selected. The levels ACPAs against these newly autoantigens 100 50 healthy controls determined enzyme-linked immunosorbent assays. Results total 135 profile included 11 antigens. Three had been reported previous studies. epitopes increased compared with those controls. Autoantibodies one antigens, lipopolysaccharide-binding (LBP), was significantly associated activities. titer LBP (anti-cLBP) closely related infection incidence Conclusion Serum technology a feasible method Anti-cLBP severity

Identification of citrullinated α-enolase as a candidate autoantigen in rheumatoid arthritis

Antibodies against citrullinated proteins are highly specific for rheumatoid arthritis (RA), but little is understood about their citrullinated target antigens. We have detected a candidate citrullinated protein by immunoblotting lysates of monocytic and granulocytic HL-60 cells treated with peptidylarginine deiminase. In an initial screen of serum samples from four patients with RA and one con...

Prohibitin as a novel autoantigen in rheumatoid arthritis

THE AIM OF THIS STUDY The aim of this study was to verify whether prohibitin is a novel autoantigen in rheumatoid arthritis. MATERIAL AND METHODS First, recombinant human prohibitin (rhPHB) protein was cloned, expressed, and purified. Then the anti-prohibitin autoantibodies were detected by western blotting by using rhPHB protein to incubate sera from patients with rheumatoid arthritis (RA). ...

Rheumatoid factor as a potentiator of anti-citrullinated protein antibody-mediated inflammation in rheumatoid arthritis.

OBJECTIVE The co-occurrence of rheumatoid factor (RF) and anti-citrullinated protein antibody (ACPA) positivity in rheumatoid arthritis (RA) is well described. However, the mechanisms underlying the potential interaction between these 2 distinct autoantibodies have not been well defined. The aim of this study was to evaluate the epidemiologic and molecular interaction of ACPAs and RF and its as...

Anti-Citrullinated Protein Antibodies as Novel Therapeutic Drugs in Rheumatoid Arthritis

We have developed a novel antibody-based treatment for rheumatoid arthritis. In a collagen antibody-induced arthritis mouse model, anti-citrullinated protein antibodies (ACPAs) prevent the onset and/or exacerbation of inflammation, and prevent or strongly reduce joint damage. For the development of this novel therapy, we focussed on rheumatoid arthritis-specific citrullinated peptide epitopes. ...

Identification of novel citrullinated autoantigens of synovium in rheumatoid arthritis using a proteomic approach

Recently, autoantibodies to some citrullinated autoantigens have been reported to be specific for rheumatoid arthritis (RA). However, an entire profile of and autoimmunity of the citrullinated proteins have been poorly understood. To understand the profile, we examined citrullinated autoantigens by a proteomic approach and further investigated the significance of citrullination in antigenicity ...
